Serine326/Threonine327/Serine328 (S326/T327/S328) is major phosphorylation site of the Hydroxycarboxylic Acid Receptor 2 (HCA2). The pS326/pT327/pS328-HCA2 antibody detects phosphorylation in response to agonists. S326/T327/S328 phosphorylation is likely to be involved in efficient ligand sequestration.

Figure 1. Agonist-induced Serine326/Threonine327/Serine328 phosphorylation of the Hydroxycarboxylic Acid Receptor 2. Upper panel, HEK293 cells stably expressing the Hydroxycarboxylic acid receptor 2 (HCA2) and transiently GRK2 (Ratio HCA2 receptor:GRK2 = 10:1) were preincubated with serum-free media. After 2 hours cells were either not exposed or exposed to 100 nM Nicotinic Acid for 30 minutes. Cells were lysed and immunoblotted with the anti-pS326/pT327/pS328-HCA2 antibody (7TM0312B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-HCA2 antibody to confirm equal loading of the gel.
